How to clear service def system message duramax.
A system tamper service bay test might clear it. Do a manual regeneration clean it up. Then it has to pass a DEF fluid quality test. If you can keep driving it, the ECM will run the tests and either clear the message or turn the light back on. It has to see the system working normal for the message to clear.

Could be the DEF pump, The DEF level sensor, The DEF injector, lines, etc. Since the check engine light is on, there will be a diagnostic trouble code set in the PCM's memory. Easiest way is to lower tank a bit to find the harness going to tank heater on top of tank. You find to wires coming out of the main def system harness leading to a two wire plug on top of tank. From the plug the wires continue to the sending unit. Unplug the plug and check resistance, if unusally high or an open is found it is bad.
